As part of the innate immune system, natural killer (NK) cells are the primary form of defense against tumor cells and assorted pathogens. Understanding the cytolytic functions of NK cells are key to understanding NK cell biology and function in adoptive immunotherapy.

NK cell cytolytic activity is often evaluated by measuring the release of radioactive chromium-51 or indium-111 from labeled tumor cells. This publication explores a non-toxic, calcein AM release assay for evaluating NK cell cytotoxicity using image cytometry.
